The findings are reported in the September 2 issue of the journal Nature. \u003cbr /\u003e\u003cbr /\u003eGleevec, it turns out, has the unique ability to bind to a protein that triggers the production of amyloid-β plaques. \u003cbr /\u003e\u003cbr /\u003eThe new research from Greengard’s lab shows that this protein, called γ-secretase activating protein (GSAP), dramatically and selectively increases the production of amyloid-β peptide, which makes up the senile plaques found in the brains of most people with Alzheimer’s. GSAP works through a mechanism involving its interactions with γ-secretase, an enzyme that chops up the amyloid precursor protein, a large molecule produced naturally in the body and found in many different types of cells. \u003cbr /\u003e\u003cbr /\u003e\u003cb\u003e“Alzheimer’s disease is a devastating disorder for which there are no satisfactory treatments,” says Greengard, Vincent Astor Professor and director of the Fisher Center for Alzheimer’s Research at Rockefeller. “Our findings reveal that γ-secretase activating protein is a potential target for a new class of anti-amyloid therapies.” Greengard won the 2000 Nobel Prize in Physiology or Medicine for research into how neurons communicate. \u003c/b\u003e\u003cbr /\u003e\u003cbr /\u003eScientists have been searching for ways to reduce amyloid-β production in Alzheimer’s patients by blocking γ-secretase, but most γ-secretase inhibitors also block the cleavage of an important immune system molecule called Notch. \u003cbr /\u003e\u003cbr /\u003eNotch plays a pivotal role in the development of blood-forming organs and the immune system. Earlier research by Greengard and his colleagues showed that Gleevec, a drug used to treat leukemia and gastrointestinal stromal tumors, successfully inhibited the ability of γ-secretase to form amyloid-β without affecting the Notch pathway. \u003cbr /\u003e\u003cbr /\u003eIn the new study, led by Gen He, a research associate in Greengard’s lab, the researchers showed that GSAP stimulates production of amyloid-β in cell lines, and that reducing GSAP reduces amyloid-β. The researchers also looked at GSAP’s action in a mouse model of Alzheimer’s disease. They knocked down the gene that codes for GSAP using RNA interference, and found that levels of amyloid-β as well as plaque development decreased. Biochemical studies showed that Gleevec reduces amyloid-β production by binding to GSAP and preventing its activation of γ-secretase. \u003cbr /\u003e\u003cbr /\u003e\u003cb\u003eUnfortunately, the Gleevec molecule does not cross the blood-brain barrier, the gatekeeper that prevents some substances in the blood from entering the brain. Based on data from a study which followed women for 35 years, this is the first research in Sweden to indicate a link between stress and dementia. \u003cbr /\u003e\u003cbr /\u003e\u003ca name\u003d'more'\u003e\u003c/a\u003e \u003cbr /\u003eThe research, published in prestigious scientific journal Brain, is based on a major population study of women from Gothenburg. A representative sample of women were examined for the first time in 1968 when aged between 38 and 60, and then re-examined in 1974, 1980, 1992 and 2000.\u003cbr /\u003e\u003cbr /\u003eA question about psychological stress was included in the 1968, 1974 and 1980 surveys and was answered by 1,415 women.\u003cbr /\u003e\u003cblockquote\u003e“Stress was defined as a sense of irritation, tension, nervousness, anxiety, fear or sleeping problems lasting a month or more due to work, health, family or other problems,” explains Lena Johansson, a researcher from the Neuropsychiatric Epidemiology Unit at the Sahlgrenska Academy’s Department of Psychiatry and Neurochemistry.\u003c/blockquote\u003e\u003cbr /\u003eDuring the 35 years of the study, 161 of the participants developed dementia, mainly in the form of Alzheimer’s disease. The risk of dementia was about 65% higher in women who reported repeated periods of stress in middle age than in those who did not. In women who reported stress in all three surveys, the risk more than doubled.\u003cbr /\u003e\u003cbr /\u003e“This is the first study to show that stress in middle age can lead to dementia in old age, and confirms similar findings from studies of animals. Stress has previously been shown to increase the risk of cardiovascular diseases such as stroke, heart attack and hypertension,” says Johansson, who also refers to earlier research at the Sahlgrenska Academy showing that cardiovascular disease can lead to Alzheimer’s.\u003cbr /\u003e\u003cblockquote\u003e“This study could result in a better understanding of the risk factors for dementia, but our results need to be confirmed by other studies, and further research is needed in the area. The precursors of these plaques are believed to be the underlying cause of the nerve cell loss that leads to the disruptions in memory that characterize Alzheimer's disease.\u003cbr /\u003e\u003cbr /\u003eThe main aim of many Alzheimer therapies is therefore to inhibit the formation of beta-amyloid. Since beta-amyloid is cleaved from the so-called amyloid precursor protein (APP), scientists have focused on stopping the two enzymes that attack the precursor protein. These act like molecular scissors and cut out the beta-amyloid fragment. Blocking these scissors precludes the formation of beta-amyloid.\u003cbr /\u003e\u003cbr /\u003eDZNE and LMU researchers have succeeded in identifying an enzyme known as alpha secretase, which cleaves the amyloid precursor protein (APP) without forming beta-amyloid. Up to this point three different candidates for this function had been under consideration, but the research team has now been able to show that the enzyme ADAM10 alone is responsible for the specific cleavage. \u003cbr /\u003e\u003cbr /\u003eDr. Stefan Lichtenthaler and his team developed highly specific antibodies that can identify the different cleavage products of the precursor protein in the brain cells of mice and in human cell cultures. Using a special technique called RNA interference, \u003cb\u003ethe researchers managed to block each of the three genes that code for the three ADAM enzymes under suspicion.\u003c/b\u003e An analysis of the cleavage products revealed that the ADAM10 gene was the only one able to prevent the formation of beta-amyloids. They confirmed their results using mass spectrometry.\u003cbr /\u003e\u003cbr /\u003e\"In ADAM10 we have identified a target molecule that plays a central role in the development of the molecular processes in Alzheimer's disease. We know that ADAM10 is less active in Alzheimer patients,\" says Dr. Lichtenthaler. Also, a more sensitive test, a PET test, will soon be available for this purpose.\u003cbr /\u003e\u003cbr /\u003eOne question that several listeners called in to discuss was whether or not they would want to know that they would soon develop Alzheimer’s. There were sensible answers on both sides of the question.\u003cbr /\u003e\u003cbr /\u003eOne teacher said that he has been teaching 30 years and is 59 years old. If he knew he would have Alzheimer’s, he would retire soon and spend his time traveling with his wife. On the other hand, if he knew he was free from Alzheimer’s, he would work until 65 because he would feel he would most likely have lots of time for travel later.\u003cbr /\u003e\u003cbr /\u003eAnother man called in whose wife was 58. She already had Alzheimer’s for many years. He said he would not have wanted to know sooner. He said it would have just made the family miserable that many years earlier, realizing what they were going to face.\u003cbr /\u003e\u003cbr /\u003eOne man said he would want to know because then he would make sure to live somewhere near a large research hospital where he could get the best care. Several other people said they would want to know so they could make the best financial decisions.\u003cbr /\u003e\u003cbr /\u003eI think it is a difficult choice. Knowing certainly does give a person more options and chances to make better decisions. However, knowing probably does rob the person of additional years that could be spent free of Alzheimer’s and its problems, fears, and indignities.\u003cbr /\u003e\u003cbr /\u003eThere is also a question of insurance. What insurance company would want to insure someone that was destined to have Alzheimer’s disease? \u003cbr /\u003e\u003cbr /\u003eWhat would you choose?
